I pinealkirtlen som også kaldes koglekirtlen (eller corpus pineale) styres døgnrytmen gennem neurotransmitteren melatonin. Men forsøg viser, at noget af denne melatonin omdannes til pinolin, som igen omdannes til DMT og 5-MeO-DMT. Da melatoninkoncentrationen i hjernen er under regulation af den mængde lys der opfanges gennem øjnene betyder det, at der er højst koncentration af melatonin om aftenen og natten, og dermed også større mængder substrat til rådighed, til videre omdannelse til DMT og 5-MeO-DMT.
